First you need either a scroll or a rune that has a magic spell inscribed on it. Scrolls are one-shot deals while you can cast a spell as many times as you like from a rune if you have the needed mana. To cast from a rune though you need to be trained in the circle of magic that rune is from. If the rune does not require a spell circle, then you can cast from it without training. An example would be a teleport rune. With those, you need no training, you just need enough mana.

Open your inventory and select the scroll or rune, a number will appear on it. Just like you can use the numbers 1 and 2 on your keyboard to switch between ranged and melee weapons, you can use the numbers that show up on the rune or scroll to select the spell to cast.
So, for example, say you want to cast Light from a scroll. Open your inventory and select the scroll. Let's say that the number that shows up on it is "4". Close your inventory and select the number 4 on your keyboard. The spell is now readied. Now, press the left mouse button and the up arrow key. You will cast the spell.

sleep spell

Related to this question: How does the sleep spell scroll work?

I followed your directions and cast the spell using a hotkey. A spark appears on my character's fingertips. How do I actually make one of these novices fall asleep?

You have to hold down the action and directional keys until your character finishes casting the spell. Also, make sure your mana is full before you do it.
